Tesamorelin is a synthetic 44-amino-acid analogue of human growth hormone-releasing factor (GRF). The peptide carries an N-terminal hexenoyl group, a structural change that distinguishes it from native GRF while retaining activity at the GRF receptor.

Tesamorelin has been studied extensively in human endocrine research, particularly in work involving growth hormone release and IGF-1 response. Peptide Grading compares current listings by supplier, quantity, stated specifications, COA availability, shipping, and price per mg.

Tesamorelin Structure

Tesamorelin retains the 44-amino-acid human GRF sequence with a hexenoyl group attached to the N-terminal tyrosine. FDA pharmacology data found greater resistance to enzymatic degradation in human serum, while receptor binding remained comparable with native GRF.

What Clinical Studies Measured

The main human studies were conducted in people with HIV-associated lipodystrophy. Their primary endpoint was visceral adipose tissue (VAT), measured by CT rather than body weight alone. IGF-1 and other metabolic measures were assessed alongside VAT.

In a 26-week trial of 412 participants, VAT decreased by 15.2% with Tesamorelin, compared with a 5.0% increase with placebo.

VAT vs Other Fat Depots

The effect was mainly seen in visceral fat. Studies did not show the same reduction in abdominal subcutaneous fat. Research also reported changes in liver fat alongside the reduction in VAT.

The VAT reduction depended on continued treatment. Participants who stopped Tesamorelin and switched to placebo showed reaccumulation of visceral fat during follow-up.

## Clinical Development

Tesamorelin was developed specifically around excess visceral abdominal fat associated with HIV lipodystrophy. Its FDA-approved indication is limited to that setting and does not represent a general weight-management indication.

## FDA Regulatory History

Tesamorelin received FDA approval for its specific pharmaceutical indication, with later formulations including EGRIFTA SV and EGRIFTA WR.

This approval applies to defined pharmaceutical formulations and should not automatically be extended to unrelated research-grade Tesamorelin materials.

Safety and Regulatory Context

Clinical studies have identified issues including IGF-1 elevation, glucose-related effects, hypersensitivity, and anti-tesamorelin antibodies. Current research materials should be assessed separately from FDA-approved formulations.

Tesamorelin and WADA

Tesamorelin is prohibited under WADA S2.2.4 as a growth hormone-releasing factor.
